實(shí)驗(yàn)室介紹

阿爾茲海默�。ˋlzheimer’s disease, AD）是最常見的神經(jīng)退行性疾病，是全球最具挑戰(zhàn)的攻堅(jiān)課題之一。AD根據(jù)臨床表現(xiàn)可以分為臨床前期和癥狀期。淀粉樣蛋白（Aβ）沉積出現(xiàn)在臨床前病理期，比癥狀期早15-20年。對(duì)比之下，神經(jīng)原纖維纏結(jié)（Tau tangle）起始于癥狀期，并與神經(jīng)元死亡和腦萎縮發(fā)生的時(shí)間高度重合。

實(shí)驗(yàn)室負(fù)責(zé)人陳曉穎博士通過系統(tǒng)分析Aβ和Tau兩種動(dòng)物模型，結(jié)合病人腦組織樣本，首次發(fā)現(xiàn)在AD的發(fā)生發(fā)展中，大腦存在固有免疫和獲得性免疫相互促進(jìn)的異常免疫應(yīng)答，打破這一異常免疫應(yīng)答中的任何一個(gè)環(huán)節(jié)將有望能延緩或逆轉(zhuǎn)AD晚期病理改變。相關(guān)工作以長(zhǎng)文形式發(fā)表于Nature，并同期被Nature 和 Nature aging高亮點(diǎn)評(píng)。這一研究揭示了神經(jīng)免疫微環(huán)境的組成特點(diǎn)及其在神經(jīng)退行疾病中的動(dòng)態(tài)演變，為理解神經(jīng)退行性疾病的發(fā)病機(jī)制提供了新的理論基礎(chǔ)，對(duì)神經(jīng)退行性病變的干預(yù)提供了全新的視角和手段。

陳曉穎，清華大學(xué)副教授，清華大學(xué)麥戈文腦科學(xué)研究院，清華-北大生命科學(xué)聯(lián)合中心，清華大學(xué)免疫所，研究員。陳曉穎博士實(shí)驗(yàn)室建設(shè)有完善的動(dòng)物模型平臺(tái)、單分子平臺(tái)和成像空間，聚焦于神經(jīng)免疫和代謝在學(xué)習(xí)記憶與神經(jīng)退行疾病中的作用機(jī)制，并探索神經(jīng)退行疾病的干預(yù)策略。

The Lab. Dr. Chen is a Principal Investigator and an Associate Professor of IDG/McGovern Institute for Brain Research at Tsinghua，Tsinghua-Peking Center for Life Science, and Institute for Immunology. Chen lab focuses on understanding the brain-immune interactions in health, aging, and disease. The major research directions in her lab are 1) decoding the immune responses in Alzheimer’s disease and therapeutics development, 2) deciphering brain-immune interface and brain-body interaction, and 3) understanding neuronal plasticity throughout AD progression.

陳曉穎博士先后在加州大學(xué)洛杉磯分校和華盛頓大學(xué)醫(yī)學(xué)院從事科學(xué)研究工作，師從華盛頓大學(xué)醫(yī)學(xué)院前神經(jīng)科學(xué)系主任Azad Bonni教授（美國(guó)國(guó)家醫(yī)學(xué)院院士， 現(xiàn)任瑞士羅氏總部副總裁）和前神經(jīng)病學(xué)系主任David Holtzman 教授（美國(guó)國(guó)家醫(yī)學(xué)院院士，前美國(guó)神經(jīng)協(xié)會(huì)主席），并和免疫系Marco Colonna 教授（美國(guó)國(guó)家科學(xué)院院士），Ken Murphy教授（免疫學(xué)教科書作者），Jony Kipnis教授（腦膜免疫的開創(chuàng)者）和哈佛醫(yī)學(xué)院再生醫(yī)學(xué)系Rudy Tanzi教授（衰老中心主任）, Jorfi Mehdi教授等有深入的國(guó)際合作。

陳曉穎博士是阿爾茲海默�。ˋD）研究領(lǐng)域的杰出青年科學(xué)家之一，其研究成果促使領(lǐng)域內(nèi)重新認(rèn)識(shí)了外周適應(yīng)性免疫在重構(gòu)疾病相關(guān)中樞炎癥以及疾病發(fā)生發(fā)展中的核心作用，并據(jù)此提出干預(yù)AD新的治療方向。陳曉穎博士創(chuàng)新性科研成果發(fā)表于Nature （2022年，2023年）等頂級(jí)期刊，受到廣泛關(guān)注。由于其研究的前瞻性、巨大的發(fā)展?jié)摿�和杰出的科研成果，陳曉穎博士收到了美國(guó)頂尖高校和研究機(jī)構(gòu)的多個(gè)offer。鑒于其學(xué)術(shù)貢獻(xiàn)，陳曉穎博士近幾年來被授予了AD研究的諸多國(guó)際大獎(jiǎng), 其中包括2024年度阿爾茨海默最有影響力研究獎(jiǎng)（AD同行選舉產(chǎn)生）；2023年阿爾茨海默協(xié)會(huì)青年成就獎(jiǎng)（封面，唯一青年P(guān)I入選），受邀在阿爾茨海默病大會(huì)做報(bào)告，代表女性科學(xué)家發(fā)言; 2022年華盛頓大學(xué)醫(yī)學(xué)院最佳神經(jīng)科學(xué)科研獎(jiǎng)；華盛頓大學(xué)醫(yī)學(xué)院阿爾茨海默研究獎(jiǎng)，神經(jīng)科學(xué)年會(huì)（Sfn）職業(yè)發(fā)展獎(jiǎng)。受Immunity（2022年，2025年）邀請(qǐng)針對(duì)神經(jīng)免疫領(lǐng)域進(jìn)行綜述和展望。相關(guān)研究還被邀請(qǐng)?jiān)贙eystone, AAI, Cell Symposium, Cold Spring Harbor Meeting 等國(guó)際大會(huì)上做報(bào)告。陳曉穎教授擔(dān)任Immunity, Alzheimer's & Dementia, Neuron, Nature Neuroscience, Science Advances, Molecular Neurodegeneration, Journal of Clinical Investigation, Cell Reports和國(guó)際阿爾茨海默會(huì)議 （Alzheimer's Association international conference，AAIC）審稿專家，Journal of Neuroimmunology和Brain Bank-The Human Brain雜志的編委。

The PI. Dr. Chen was a faculty member in the Department of Neurology, Hope Center for Neurological Disorders, Knight Alzheimer’s Disease Research Center at Washington University School of Medicine. She has contributed significant insights into understanding neuronal plasticity during development (2017, Journal of Neuroscience; 2016 & 2020, Protein & Cell), learning activities (2022, Nature), and neuron-immune interactions in neurodegenerative disease (2022, Immunity; 2023, Nature). She has received numerous honors and awards, including the Jeffrey L. Morby Prize for Best Research in Alzheimer’s Disease from the Cure Alzheimer’s Fund (2024), the Early Career Achievement Award from the Alzheimer’s Association (2023), the Career Development Award from the Society for Neuroscience (2023), the Coins for Alzheimer’s Research Trust Award from the Rotary Foundation (2023), the O’Leary Prize for Best Neuroscience Research at Washington University (2022), the Poletsky Award for Alzheimer’s Disease Research from Washington University (2022), and the Tau Consortium Fellowship from the Rainwater Charitable Foundation (2019). She is a member of Society for Neuroscience, Alzheimer’s Association, American Society for Neurochemistry, American Neurological Association and International Society for Molecular Neurodegeneration. Dr. Chen has reviewed papers for journals, including Immunity, Nature Neuroscience, Alzheimer's & Dementia, Science Advances, Cell Reports, Molecular Neurodegeneration, Journal of Clinical Investigation, Journal of Neuroimmunology (Editorial board member), and Translational Neurodegeneration.
