一、實(shí)驗(yàn)室介紹

張翼研究員在北京大學(xué)獲得學(xué)士和博士學(xué)位,已加入首都醫(yī)學(xué)科學(xué)創(chuàng)新中心(CIMR)創(chuàng)建獨(dú)立實(shí)驗(yàn)室。張翼實(shí)驗(yàn)室的主要研究興趣是開發(fā)新型計(jì)算和實(shí)驗(yàn)方法,從靜態(tài)、離散的樣本中準(zhǔn)確重建出動(dòng)態(tài)、連續(xù)的生物學(xué)過程的歷史軌跡,以便更好地理解發(fā)育、衰老、疾病發(fā)生發(fā)展的分子機(jī)制,為重大疾病的診斷和治療提供理論依據(jù)與技術(shù)支撐。

本實(shí)驗(yàn)室的工作特點(diǎn)為干濕結(jié)合、學(xué)科交叉。歡迎不同背景的研究同事加入團(tuán)隊(duì)。

二、實(shí)驗(yàn)室研究方向

實(shí)驗(yàn)室將綜合利用深度學(xué)習(xí)、生物信息學(xué)、分子生物學(xué)、細(xì)胞生物學(xué)、動(dòng)物模型、單分子測(cè)序、單細(xì)胞測(cè)序等技術(shù)和手段,開展(不限)以下課題研究:

1.開發(fā)前沿計(jì)算工具,即新型計(jì)算生物學(xué)方法,用于解析復(fù)雜數(shù)據(jù)中隱含的動(dòng)態(tài)特征信息。

2.開發(fā)新型單細(xì)胞、單分子水平的實(shí)驗(yàn)和計(jì)算方法,從靜態(tài)樣本中重建生物過程的動(dòng)態(tài)歷史,鑒定其中關(guān)鍵的分子事件。

3.綜合運(yùn)用正向遺傳學(xué)篩選、反向遺傳學(xué)操作、單分子測(cè)序、單細(xì)胞多組學(xué)分析手段,解析表觀基因組復(fù)制的分子機(jī)制,特別是在衰老、疾病中表觀遺傳信息復(fù)制的分子機(jī)制。

1.開發(fā)從單細(xì)胞ATAC數(shù)據(jù)中計(jì)算細(xì)胞分裂年齡的方法 (Nat.Biotech., 2024)

2.發(fā)現(xiàn)DNA二級(jí)結(jié)構(gòu)影響表觀基因組復(fù)制 (BioRxiv, 2024)

3.開發(fā)將神經(jīng)活動(dòng)轉(zhuǎn)化為遺傳可操作對(duì)象的方法 (J.Neurogenetics, 2012)

01、科研助理

1)崗位職責(zé)

1.獨(dú)立或協(xié)助團(tuán)隊(duì)開展課題研究;

2.協(xié)助實(shí)驗(yàn)室運(yùn)行管理。

2)任職要求

1.大學(xué)本科學(xué)歷及以上,生物學(xué)、生物信息學(xué)、演化生物學(xué)及其它相關(guān)專業(yè);

2.掌握分子生物學(xué)操作及原理,有二代測(cè)序、三代測(cè)序、細(xì)胞生物學(xué)、模式動(dòng)物等研究經(jīng)歷優(yōu)先;

3.具備良好的溝通能力和團(tuán)隊(duì)協(xié)作能力;

4.具備良好的英語讀寫和日常交流能力。

02、博士后

1)崗位職責(zé)

1.以科學(xué)研究工作為主,在PI指導(dǎo)下獨(dú)立開展研究工作,按計(jì)劃完成博士后研究任務(wù);

2.與合作導(dǎo)師共同承擔(dān)科研課題,發(fā)表高水平研究論文,申請(qǐng)各類科研項(xiàng)目、專利,協(xié)助指導(dǎo)研究生工作。

2)任職要求

1.具有生命科學(xué)、生物信息學(xué)、計(jì)算機(jī)科學(xué)、物理學(xué)或其他相關(guān)領(lǐng)域的研究背景,在國(guó)內(nèi)外已獲得或即將獲得博士學(xué)位(原則上不超過3年),有物理、數(shù)學(xué)、計(jì)算機(jī)、生物信息學(xué)訓(xùn)練背景或三代測(cè)序、單細(xì)胞測(cè)序、Perturb-seq實(shí)驗(yàn)背景者優(yōu)先考慮;

2.以第一作者身份在領(lǐng)域內(nèi)主流期刊上發(fā)表過或即將發(fā)表研究論文(在BioRxiv上已公開),能夠獨(dú)立開展研究工作;

3.具備獨(dú)立開展科研工作和指導(dǎo)研究生能力,具有嚴(yán)格的學(xué)術(shù)道德、高漲的科研熱情、嚴(yán)謹(jǐn)?shù)目蒲袘B(tài)度、良好的溝通能力和團(tuán)隊(duì)合作精神,具有誠(chéng)實(shí)健康、樂觀堅(jiān)強(qiáng)、有上進(jìn)心、勤勞刻苦的個(gè)人品質(zhì);

4.具有高效的信息檢索、英語文獻(xiàn)閱讀、論文撰寫能力,具備應(yīng)有的分子生物學(xué)或生物信息學(xué)基本技能,熟知技術(shù)背后原理。

Laboratory Introduction

The Zhang laboratory develop novel computational and experimental methods to track human development, aging, and disease across time domains on single-cell, single-molecule resolution.

The laboratory conducts cross-disciplinary research with a combination of computational and experimental biological methods.We welcome colleagues with background in physics, computer science, biology, bioinformatics, and medicine to join our team.

Major Research Projects

The laboratory uses a combination of deep learning, bioinformatics, forward and reverse genetics, and sequencing approaches on human samples and animal models, to:

a.Develop methods to extract latent information from biological data.

b.Develop novel methods to resolve dynamic biological process from static samples.

c.Dissect the molecular mechanism of epigenome replication, particularly in aging and disease.

Major Contributions

a.Developed method to decode cell replicational age from single-cell ATAC-seq data (Nat.Biotech., 2024)

b.Provided evidence for how DNA secondary structure affect epigenome replication (BioRxiv, 2024)

c.Developed tools to convert Drosophila neuronal activity into genetic manipulable object (J.Neurogenetics, 2012)

Research Assistant

Main Responsibilities

a.Conducting research projects either independently or in a team.

b.Assisting PI in lab routine management.

Qualifications

a.B.S.or higher degree in biology, bioinformatics, evolutionary biology or other relevant majors is required.

b.Research experience in bioinformatics, next-generation sequencing, single-molecule sequencing (Nanopore or PacBio), cell biology and/or model animal works is preferred.

c.Self-motivated researcher with good communication skill and teamwork ability.

d.Proficient in English reading, writing, and daily communication.Passed CET 6.

Postdoctoral Researcher

Main Responsibilities

a.Conducting research projects independently.

b.Assisting PI to mentor graduate students, and write grant proposals and manuscripts.

Qualifications

a.Backgrounds in computer science, physics, bioinformatics, biology or biomedical engineering.Either about obtaining a Ph.D.or recently obtaining a Ph.D.degree (ideally within the last 3 years).

b.Have first author paper(s) published in (or accepted by) reputable journals in the field.

c.Have the ability to independently carry out scientific research and guide graduate students, have scientific research enthusiasm and rigorous scientific research attitude.

d.Learns quickly.

e.Strong research experience in either:

 (i) Perturb-seq, single-cell sequencing, single-molecule sequencing, epigenomics.

 (ii) Bioinformatics, deep learning, algorithm development.

f.Proficient in English reading, writing, and daily communication.
